Abstract: Store root (SR) formation in cassava is an important trait related to starch formation and quality. It is the result of secondary growth and starch deposition in parenchyma cells. No distinct developmental stage for this organ can be established based on root morphology ony because of indeterminate SR growth nature and the continuous cambium activities which give raises to the root diameter. In order to understand cassava store root development our research is focusing on the pattern of tissue differentiation during the raising of diameter. Store root morphology (root diameter), anatomy and deferential gene expression are being studied. Changing in root diameter was, first used to differentiate stages of SR formation. Anatomical observation with different tissue stains, revealed the same tissue pattern in five SR diameter, except the fibrous root. Cambium activity and radial dislocation were observed as SR were being formed. Proportion of different specialized cell layers such as store parenchyma, cabium and cortex varied with the raise of SR diameter. An SDS-PAGE analysis of protein extracted from SR of five different diameter indicated a simple protein pattern and a differential pattern between fibrou's root and store root. Later SR stage also showed particular pattern differing from the early ones. High resolution 2-D gel electrophoresis analysis separated several classes of proteins related to SR diameter. These results are discussed in relation to the store root anatomical changes during the secondary growth.
